Can I appoint substitute trustee(s) who will only replace another specific trustee?

Based on the platform, the default clause is for the substitute trustee(s) to replace ANY of the testamentary trust's trustee(s) should they die before you, is unable or unwilling to serve or continue to serve.

Nevertheless, if say for example you have joint trustees A and B but want C, D or E to only replace A while X, Y or Z to only replace B, 

you can do so by keying in the following under "Additional Trust Instructions" in the testamentary trust page:

Should {Relationship}, {Name}, {ID no.}, who is the {Trust Name} Trust's Trustee die before me, is unable or unwilling to serve or continue to serve, I specifically appoint the following personnel in order of priority to replace him/her and ONLY him/her as one of the joint trustees of the {Trust Name} Trust:

(a) {Relationship}, {Name}, {ID no.}
(b) {Relationship}, {Name}, {ID no.} ....
Should {Relationship}, {Name}, {ID no.}, who is the {Trust Name} Trust's Trustee die before me, is unable or unwilling to serve or continue to serve, I specifically appoint the following personnel in order of priority to replace him/her and ONLY him/her as one of the joint trustees of the {Trust Name} Trust:
(a) {Relationship}, {Name}, {ID no.}
(b) {Relationship}, {Name}, {ID no.} ....

Following the example above:

Remember to leave the substitute trustee(s) section blank if you intend for the substitution to go according to this instead.
